Pyruvate kinase, a glycolytic enzyme, facilitates the conversion of phosphoenolpyruvate to pyruvate. In mammals, the M1 isoform (PKM1) is prevalent in most adult tissues (1), while the M2 isoform (PKM2), an alternative splice variant of M1, is expressed during embryonic development (1). Cancer cells uniquely express PKM2 (1-3), which is crucial for aerobic glycolysis in tumors, the Warburg effect (1). When cancer cells shift from the M2 to the M1 isoform, aerobic glycolysis decreases, oxidative phosphorylation increases, and tumorigenicity in mouse xenografts is reduced (1). However, recent studies suggest that PKM2 is not essential for all tumor cells (4), being active in non-proliferative and inactive in proliferative tumor cell populations in the studied tumor model (4).
